Abstract:

The detection of trace components within complex systems of traditional Chinese medicine (TCM) has long represented a formidable and significant challenge in the realm of TCM analytical research. This study proposes a novel approach that combines deep eutectic solvents (DES) with dispersive liquid–liquid microextraction (DLLME) to simultaneously extract, enrich, and quantify trace flavonoid components derived from Ophiopogon japonicus in anti-aging oral liquid. Based on a univariate experimental design, the optimal extractant (dichloromethane) and dispersant (choline chloride:1,4-butanediol, 1:3) were selected. Additionally, a hybrid analytic hierarchy process (AHP)-criteria importance through intercriteria correlation (CRITIC) weighted approach was employed to assign weights to the evaluation indicators, including methylophiopoganonone A (MOA) and B (MOB). Significant variables were screened and optimized using Plackett–Burman and Box–Behnken designs. Under optimal conditions, the HPLC-DAD quantitative analysis, which combined the enrichment capabilities of DLLME and solid phase extraction (SPE), demonstrated excellent performance, with enrichment factors for MOA and MOB of 96 and 90, respectively. The linear range was 0.089–253.75 μg mL?1 (r2 ≥ 0.9999), with the limits of detection (LOD) and quantification (LOQ) of 0.027 μg mL?1 and 0.091 μg mL?1, respectively. The recovery rates ranged from 87.55 % to 103.84 %. Furthermore, the approach was successfully applied to determine the concentrations of two characteristic flavonoids in ten batches of anti-aging oral liquid. The developed method is simple, rapid, cost-effective, and requires minimal solvent usage, as well as achieving high enrichment factors. Moreover, it can provide valuable reference and ideas for the enrichment and quantification of trace components in TCM prescriptions.
